Title: CYTOKINE EXPRESSION IN LYMPH NODES INFECTED WITH MYCOBACTERIUM BOVIS

Technical Abstract: Retropharyngeal lymph nodes were taken from animals 6 and 8 weeks after instillation of 4.8 x 10*5 Mycobacterium bovis into the crypts of the palatine tonsils. Tissues were routinely processed into paraffin blocks and sections were probed for the presence of tumor necrosis factor (TNF)-al interleukin (IL)-10, and gamma-interferon (gamma-IFN) mRNA using digoxigenin-labeled riboprobes in an in situ hybridization procedure. In infected animals, TNF-alpha was present in abundant quantities in macrophages surrounding granulomas, however there was no TNF-alpha in epithelioid macrophages or multinucleate giant cells. IL-10 was present in lymphocyte-like cells, often in the compressed lymphoid tissue adjacent to a granuloma. Staining for gamma-IFN was very weak.
