Technical Abstract: Beta-1,3-glucanases are involved in the breakdown and turnover of beta-1,3- glucans, which are found in several plant organs. The beta-1,3-glucanases are critical for normal microsporogenesis in that they degrade callose walls surrounding the microspore tetrads. They also are frequently associated with a role in plant defense, and first were described as belonging to the pathogenesis-related family of proteins which accumulate rapidly in response to pathogen attack or stress. Oligonucleotide primers designed for conserved sequences from coding regions of beta-1,3-glucanase genes from diverse taxa were used to amplify related sequences from soybean. These amplified sequences were cloned and grouped into at least thirteen classes of beta-1,3-glucanase which did not cross-hybridize under stringent hybridization conditions. Genetic mapping of these thirteen classes is being conducted to locate their positions within respect to soybean linkage groups, and their possible association with both male- sterile and disease-resistant loci. So far, two have been shown to co- segregate with the fungal-resistant beta-1,3-glucanases previously isolated from pea and soybean.
