Technical Abstract: Beta-1,3-glucanases play an important role during microsporogenesis in that they aid in the breakdown of the callose walls surrounding the microspore tetrads. This breakdown allows their continued development. Beta-1,3- glucanases also are frequently associated with a role in plant defense. They were first described as belonging to the pathogenesis-related family of proteins which accumulate rapidly in response to pathogen attack or stress. In our study we used oligonucleotide primers for conserved sequences from coding regions of beta-1,3-glucanase genes obtained from several unrelated plant taxa to amplify related sequences in soybean. These amplified sequences were cloned and grouped into at least ten classes of beta-1,3-glucanase which did not cross-hybridize under stringent hybridization conditions. The positions of these nine classes of beta-1,3- glucanase are being genetically mapped within already established soybean linkage groups to determine their relationship with known resistance and male-sterility genes.
