Title: Mapping of Microsatellite Markers Developed from Swine Chromosome 6 Enriched Libraries

Technical Abstract: Swine chromosome 6 enriched libraries have been produced using both flow cytometric and microdissection techniques. These libraries were successfully used to develop new chromosome 6 microsatellite markers. We targeted swine chromosome 6 because the gene responsible for Porcine Stress Syndrome (PSS), the calcium release channel (CRC) gene, has been mapped to this chromosome, and this region is also known to contain a QTL which beneficially affects a variety of carcass and production traits. Purity of the chromosome populations used to generate the libraries was verified by fluorescence in situ hybridization (FISH) analysis using painting probes generated from the isolated products. The libraries were screened with dinucleotide repeat probes and a total of 132 unique (CA)n repeat containing clones were isolated. Thirty of these were suitable for genotyping the University of Illinois swine reference families and the results of linkage analysis (CRIMAP 2.4; LOD > 3.0) showed that 24 of the microsatellites mapped to swine chromosome 6. Five of the new markers map to a region on the q arm that currently has only two markers over a 40 cM area, and three more of the markers are within 3 cM of the CRC locus. Several other markers, because of their PIC values, should provide more informative markers in other areas of the map. Most of the markers can also be used fluorescently for automated genotyping. This set of 24 new chromosome 6 markers will therefore be useful in helping define QTL associated with swine chromosome 6.
