Title: The Influence of the Conceptus and Early Progesterone Treatment on Uterine Protein Secretion in Swine

Technical Abstract: White crossbred gilts with a normal estrus cycle were heat checked twice daily and were assigned at estrus to treatment groups: 1) cyclic-control, 2) pregnant-control, 3) cyclic-progesterone (P4) and 4) pregnant-P4 treatment. Gilts receiving P4 were given 200 mg i.m. at 48 and 72 h post-es Gilts from each treatment group were slaughtered on d 10, 11, 12, 13 or 15 (n=3 to 6 for each treatment day). At slaughter, the uterus was collected, flushed with 20 mL .1x leucine minimal essential medium (MEM)/horn and endometrium was dissected and cultured for 24 h in MEM plus 50 uCi 3H-leucine. Uterine flushings were assayed for protein, retinol binding protein (RBP), acid phosphatase (AP), uteroferrin (UF) and oxytocin (OT). Endometrial cultures were assayed for nondialyzable radioactivity (NDR), AP, and RBP. Each protein in the uterine flushings increased severalfold from d 10 to 13 and did not change between d 13 and 15. Total protein did not differ between status or P4 treatment groups. Analysis indicated a difference between pregnant and cyclic gilts for UF and OT (P<.01). Both UF and OT were greater in uteri of pregnant gilts than cyclic gilts from 288 to 312 h post-estrus (d 12 and 13). Analysis also indicated a difference between control and P4-treated gilts for uterine flush RBP (P<.01), UF (P<.01) and AP (P<.01). Both UF and RBP were greater in P4-treated gilts from 240 to 264 h post-estrus (d 10 and 11) compared to control gilts. AP secreted in culture was greater in P4-treated gilts (main effect; P<.05). No effect of status or treatment was obtained for NDR or RBP in culture. These data suggest that 1) OT and UF may be controlled by conceptus estrogen, 2) RBP and UF may be controlled by a set period of P4 influence.
