Title: IN VITRO AND IN VIVO REGULATION OF IL-I RECEPTOR MRNA IN BOVINE LEUKOCYTES

Interpretive Summary:

Technical Abstract: Previous studies have shown that IL-1 receptors on human cells are regulated in response to numerous mediators, including cytokines and glucocorticoids. Information on IL-1 receptors on large animal species has been lacking. In this study, we examined the effects of various stimuli on the in vitro regulation of IL-1 receptors in bovine PBMCs and PMNs, and the effects of in vivo treatment of calves with dexamethasone on the ex vivo expression of IL-1 receptors. Our results indicate that IL-1RI and IL-1RII gene expression in PBMCs was increased by in vitro incubation with rBoIL-1beta, rBoGM-CSF, rBoIL-4 and dexamethasone. These same stimuli up-regulated IL-1RII expression in PMNs. rBoIFN-gamma down-regulated the expression of IL-1RII and up-regulated the expression of IL-1RI in bovine PBMCs. In contrast, IL-1RII was up-regulated by rBoIL-1beta and IL-1RI was down-regulated by rBoIL-1beta and rBoIFN-gamma in PMNs. These data suggest that IL-1RII and IL-1RII are regulated differently in bovine leukocytes and that gene expression is different for neutrophils and mononuclear cells. We went on to show that in vivo treatment of calves with dexamethasone also up-regulated IL-1RI and IL-1RII mRNA in bovine PBMCs and PMNs. IL-1RII mRNA was strongly induced in both PBMCs and PMNs at 24 hours, but not 72 hours, after dexamethasone injection.
