Title: Analysis of Differently Expressed Mrna Between Virulent and Attenuated Mdv-1 in Cef by Differential Display

Technical Abstract: During the process of attenuating Marek's disease virus (MDV) by passage in tissue culture, several changes in the virus genome have been reported. However, it is not clear which changes(s) on the genome is responsible for the attenuation. Differential display (dd) is a novel technique which utilize RT-PCR with short 5' primers and anchored oligo-dT 3' primers to compare expressed mRNA in two different cell populations. We have applied this technique to the RNA from virulent and cell culture-passaged attenuated Md11-infected chicken embryo fibroblasts in order to detect differences in mRNA expression in these cells. With 132 reactions, we found four virulent virus-specific bands. They were localized near the 132bp repeat region, on ICP4 gene region, on VP16 gene region and on the BamHI-D fragment, respectively. However, the difference in expression was confirmed by Northern blot only with the dd fragment located near the 132bp prepeat region.
