Technical Abstract: Molecular Hybridization is the term used to describe the detection of specific DNA sequences after their electrophoretic separation. Many variations on the original procedure developed by Southern have been described in the scientific literature. Because of this profusion, it is often difficult for a inexperienced person to know which subsets of the various versions are compatible. Our goal in this chapter is to provide a complete set of protocols of each of the steps in molecular hybridization that are known to work together. They are presented among with commentaries on each step and when appropriate, discussions of allowable variations.
