Title: Evidence of Thinopyrum Junceiforme DNA Fragments in the Modified Durum Wheat Genome Utilizing Rapd and Rflp.

Technical Abstract: Wild grass species of the tribe Triticeae are an excellent reservoir of agronomically important traits. Intergeneric crosses between durum wheat and grass species provide an opportunity to: 1) transfer important traits into durum; 2) generate valuable research material that could be used to study genome relationships among different species of the Triticeae; and 3) develop molecular markers that are species-specific or alien chromatin-specific. Intergeneric hybrids were made between durum wheat (cultivars Langdon, Lloyd and Durox) and Thinopyrum junceiforme. RAPD 9-mer oligonucleotides and RFLP techniques were applied to identify alien DNA fragments in the hybrid genome. Several amplified DNA fragments from Th. junceiforme were identified in the hybrid durum genome using primers PR23 and PR41. These alien DNA products (for example, 1.7 kb fragment) were also detected in sterile and fertile BC1 generations of Lloyd x Th. junceiforme through Southern analysis. Similarly, genomic DNA restricted with Hind III and Eco RI and probed with either genomic DNA or amplified DNA with PR41 of Th. junceiforme showed the presence of alien DNA in the hybrid genome of durum wheat. Species-specific markers of durum wheat and grass species, pairing among parental chromosomes, and presence of alien DNA in the modified durum wheat genome will be discussed.
