Title: MAP OF A SINGLE LOCUS IN ARABIDOPSIS THALIANA THAT CONFERS TOLERANCE TO TOBACCO RINGSPOT VIRUS INFECTION

Technical Abstract: We previously reported the identification of a locus, TTR1, in Arabidopsis thaliana that confers tolerance to tobacco ringspot virus infection. Initial mapping studies employing 17 PCR-based markers dispersed across the Arabidopsis genome and 40 F3 families derived from a cross of a sensitive line, Estland, and a tolerant line, Col-0, indicated that the TTR1 locus is positioned on the bottom arm of chromosome V between nga76 and LFY3 with recombination rates of 33% and 15%, respectively. Subsequently, several other PCR-based markers including, nga129, ve027, and DFR, have been used to map the locus in 295 F3 families, 143 F3 families, 112 F3 families, respectively. Mapping data on these F3 families suggested that the locus is between DFR and ve027 with genetic distances of 6 cM and 5 cM, respectively. Markers between DFR and ve027, such as cra1 and CIC11H2, YUP5F5 are being used to more precisely map the locus.
