Technical Abstract: The humoral and cell mediated immune response to glycoproteins gB, gC, and gD of pseudorabies virus (PRV) was investigated by vaccinating pigs with recombinant vaccinia viruses expressing these glycoproteins. Peripheral blood mononuclear cells from vaccinated pigs were isolated to examine the in vitro lymphocyte proliferative response to PRV antigen, and sera from these pigs were tested for virus neutralizing antibodies to PRV. Pigs vaccinated with the recombinant gD vaccine responded with high virus neutralizing antibody titers but low lymphocyte proliferative responses to PRV, while pigs vaccinated with the recombinant gB vaccine had low virus neutralization titers but high lymphocyte proliferative responses. Pigs vaccinated with the recombinant gC vaccine had neither significant virus neutralization titers nor lymphocyte proliferative responses. These results indicate important roles for gD in humoral protection and gB in cell mediated protection to PRV infection. In addition, results of current research to further determine the nature of the proliferative response will be presented.
