Technical Abstract: The construction of species-specific comprehensive genomic maps is facilitated when anonymous polymorphic markers have also been physically anchored. This allows both marker assignment and orientation, and ultimately indicates chromosomal coverage. Although major advances have been made in the development of a porcine microsatellite linkage map, the number of microsatellite loci physically anchored to individual porcine chromosomes is much lower than other species. To physically anchor an unassigned porcine microsatellite linkage group, a cosmid containing a microsatellite known to be near one end of the group (Sw2415) was used for fluorescence in situ hybridization. To confirm the assignment and orient the linkage group, another cosmid containing the microsatellite Sw2419 was also anchored using fluorescence in situ hybridization.
