Title: ASSOCIATION OF ALLANTOIC FLUID AND ENDOMETRIAL RETINOL BINDING PROTEIN (RBP) AND UTEROFERRIN (UF) DURING GESTATION IN THE PIG

Technical Abstract: Uteroferrin in the presence of ascorbic acid catalyzes lipid peroxidation in cell membranes of reproductive tissues and RBP inhibits this reaction. An association between endometrial and allantoic fluid RBP and UF throughout gestation is likely if RBP serves a protective role. Twenty white crossbred gilts were mated at estrus (d 0) and slaughtered on d 25, 30, 40, 60, 70, or 90 (n = 3 or 5/d). After slaughter, allantoic fluid wa pooled from each gilt for RBP RIA and UF (acid phosphatase; AP) analysis. Endometrium was collected for 1) RBP mRNA slot blot analysis and 2) 24-h culture (500 mg of tissue, 15 ml Eagle's MEM + 50 uCi 3H-leucine) to measure production of protein, RBP, and AP. Allantoic fluid protein, RBP and AP concentration and total content increased (P < .05) from d 25 to 40 and RBP and AP were correlated (r = .6; P < .01) throughout gestation. Endometrial culture RBP and AP increased during early (d 25 to 40) and late e(d 70 to 90) stages of gestation and were correlated (r = .4; P < .05). Thus, results indicate an association between RBP and UF throughout pregnancy. This data is consistent with a proposed role for RBP in protecting against UF-induced lipid peroxidation, particularly from d 25 to 40.
