Interpretive Summary: This manuscript summarizes all of the results of an international workshop conducted over the last 3 years to compare and standardize the reactivities of monoclonal antibodies (mAb) that recognize pig leukocyte cell surface markers; this was sponsored by the International Union of Immunological Scientists. A total of 136 mAb were provided by scientists in government, university and commercial institutions. Altogether scientists in 29 laboratories in 14 countries participated in the Workshop . All mAb were compared for their binding to cells from different pig tissues (blood, spleen, lymph nodes, bone marrow), to purified T, B, and macrophage cell populations and to cultured pig cell lines. Tests were performed in several independent lab oratories to verify whether certain mAb should be assigned specific "clusters of differentiation" antigen numbers (CD#) or Swine Workshop Cluster Numbers (SWC#). Clustered mAb were then directly compared to determine whether they bound to the same site, or epitope, on the CD or SWC antigen to establish how different the individual mAb were. As a result of this international workshop 62 mAb have been given CD#s or SWC#s. This designation will enable scientists worldwide to compare results with these standardized mAb as they assess swine immune responses to infectious agents, vaccines, transplants, etc. A second workshop started in Spring, 1993.
Technical Abstract: The First International Swine Cluster of Differentiation (CD) Workshop was organized to standardize the assignment of mAb reactivity to swine leukocyte antigens, using the nomenclature adapted for the human CD antigens. This workshop took place over the last three years with the summary meeting being held in Budapest, Hungary, on August 16, 1992 and was ssupported by the Veterinary Immunology Committee of the International Unio of Immunological Scientists. A short summary of the results of this workshop has been published (Lunney, 1993). This manuscript presents the characteristics of the full panel of mAb offered for the workshop and highlights the mAb that were officially designated as reactive with specific swine CD antigens or with SWC antigens as a result of this workshop. The manuscripts in the second half of this volume report the detailed data supporting these assignments. Tests of cell/tissue specificity were performed on each section's panel of mAb, and new clustering analyses conducted. As a result of these tests, CD# or Swine Workshop Cluster # (SWC#) were assigned to 62 mAb with confirmed subset reactivities; epitope analyses were then performed on most of these important mAb. This workshop has enabled scientists to definitively establish a panel of mAb reactive with swine CD molecules for the first time. It is clear, from data in other species, that many new mAb and CD assignments are required for future progress, especially for B and myeloid cell subsets, and for activation and adhesion molecules. This First Swine CD Workshop has set the stage for future research and productive interactions between swine immunologists as well as set the basis for the Second Swine CD Workshop.
