Interpretive Summary: This manuscript summarizes the results of part of the second round of analyses performed by scientists participating in the First International Swine CD Workshop. This workshop was set up to compare and standardize the reactivities of monoclonal antibodies (mAb) that recognize pig leukocyte cell surface markers to assign them specific cluster of differentiation numbers (CD#). For the second round analyses, the reactivities of 54 mAbs on T cell subsets or lines were analyzed by flow cytometry a nd the data subjected to statistical clustering to identify groups of mAbs that reacted similarly with specific pig T cells. These results confirmed the assignment of 5 monoclonal antibodies (mAb) to the CD2 T cell cluster. Another mAb, MSA2, originally clustered with these 5 CD2 specific mAb but was not assigned to a CD grouping because of discordant binding results and the lack of biochemical data. Clustered mAbs were examined in more detail to determine whether they bound to the same area, or epitope, of the CD antigen. Epitope analyses and the molecular weight of the precipitated antigen confirmed that 3 mAb (MSA4, MAC80, MAC83) bound to the same epitope, CD2a, of the swine antigen. Two other mAb (1038-8 and PG168) appear to bind t he same epitope but their antigen molecular weight has not been confirmed; they are noted as reacting with the wCD2a antigen. These results will enable researchers to readily compare results of their studies using porcine CD2 specific mAb and thus enrich swine immunologic studies.
