Interpretive Summary: Soybean fatty acid desaturase 8 (GmFAD8) is a nuclear encoded gene whose protein product is transported into the chloroplast membranes. We sequenced the genomic FAD8 gene in order to characterize gene structure and to determine FAD7 gene expression in response to cold stress and pathogen infection of soybean leaves. The genomic sequence of soybean GmFAD8 (2480 nucleotides) was determined and reported to GenBank and was given the accession number HM769341. The structure and deduced amino acid sequence of soybean FAD8 is similar to other higher plant plastidal omega-3 desaturases: 8 exons and 7 introns, predicted proteins of 453 amino acid residues containing three conserved histidine box motifs, amino terminal chloroplast transit peptides, and a molecular mass of 51.4. GmFAD8 has 76% amino acid sequence identity to GmFAD7.

Technical Abstract: The genomic sequence of soybean GmFAD8 (2480 nucleotides) was determined and reported to GenBank and assigned the accession number HM769341. The structure and deduced amino acid sequence of soybean FAD8 is similar to other higher plant plastidal omega-3 desaturases: 8 exons and 7 introns, predicted proteins of 453 amino acid residues containing three conserved histidine box motifs, amino terminal chloroplast transit peptides, and molecular masses 51.4 kDa. GmFAD8 has 76% amino acid sequence identity to GmFAD7. GmFAD8 exons also range in size from 67 to 521 nucleotides and the 7 introns also range in size from 90 to 393 nucleotides. Two complete copies of GmFAD8, one on chromosome 3 and one on chromosome 1 of the Williams 82 soybean genome were found with strong sequence similarity to GmFAD8 of cultivar Dare. Comparison of the Dare GmFAD8 deduced peptide sequence to published soybean FAD8 peptides revealed 94 % identity to the homologous sequences.
