Interpretive Summary:

Technical Abstract: One former commercial variety, EL-A012206 (ACH185), and the pollen parent for the commercial variety USH20, EL-A015030 (SP7622) were germinated in H2O and 0.3% H2O2. Samples were collected at 0, 3, 6, 12, 18 and 24 hours of treatment for both varieties. RNA was extracted from the tissue and RT-PCR was performed using 343 primers developed from putative genes related to development and stress. Expression patterns determined by RT-PCR were qualified between varieties and treatments and k-means clustered into patterns of similar expression. Five clusters of expression patterns were shared between both varieties and treatments. In the good emerger, one cluster, 9, was shared in SP7622 between the H2O and H2O2 treatment. No clusters were shared solely between the two ACH185 treatments and only one cluster, 7, was limited to the H2O treatment of the low vigor variety (ACH185). This cluster, characterized by expression at 1 h of treatment and little expression at later time points, suggests that the timing of gene expression may be playing a role in reducing germination vigor. Between the two varieties and treatments only 16 genes shared expression patterns in the H2O2 treatment. These genes shared between the two vigor treatments are potential molecular markers for hydrogen peroxide induced vigor.
