Technical Abstract: In this chapter, methods for the isolation of yeasts and their maintenance are discussed as well as procedures for morphological and physiological characterization of strains. In an era of microbial strain identification by gene sequencing, some will ask why there is a need for morphological and physiological tests, especially considering that identification by gene sequencing is faster and more accurate.
