Interpretive Summary:

Technical Abstract: The production of hybrid-onion seed is dependent on cytoplasmic-genic male sterility (CMS) systems. For the most commonly used CMS, male-sterile (S) cytoplasm interacts with a dominant allele at one nuclear male-fertility restoration locus (Ms) to condition male fertility. We are using a proteomics approach to study differences for mitochondrial proteins in male-sterile (S msms) versus restored male-fertile (S Msms) plants. We isolate mitochondria from both populations, extract mitochondrial proteins, and run 2D gels. Unique spots are subjected to tandem mass spectrometry to produce partial sequences from proteins located in mitochondria and to identify matches for these oligopeptides. One unique spot showed similarity to aconitate hydratase and glycine dehydrogenase, both nuclear-encoded proteins that can be imported into the mitochondria. Nuclear polymorphisms are being identified and their linkages to Ms assessed.
