Technical Abstract: Seasonal Ornithobacterium rhinotracheale (ORT) infection of chickens and turkeys causes significant losses to the poultry industry. Little information is available in the literature on the virulence factors of ORT. This study describes a hemolysin-like activity of ORT strains, as observed on sheep blood agar and by in vitro hemolysis assays. iTRAQ (isobaric tagging for relative and absolute quantification) labeling and mass spectrometry sequencing of peptide fragments revealed four peptide sequences that were identical to the corresponding Mannheimia haemolytica LKTA peptide sequences. Results from Western blots with monoclonal antibodies against M. haemolytica LKTA showed weak reactions with two proteins at molecular masses of 181,000, 102,000 and a strong reaction with a band of 56,000. The 56,000 kDa band is consistent with an IgG binding protein. Identification and confirmation of identity of these proteins by mass spectrometry sequencing is in progress. This is the first report of a hemolysin-like activity of ORT.
