Technical Abstract: Recent work in our laboratory has focused on development of LC methods with diode array and/or mass spectrometry (ms) detection for the simultaneous determination in supplement tablets and fortified foods of several water-soluble vitamins (WSV) including: thiamin, niacin, pyridoxine, pantothenic acid, folic acid, biotin, riboflavin, and ascorbic acid. As part of a collaborative effort with NIST to characterize the new SRM 3280 Multivitamin Tablets, isotopically labeled standards for four WSV were obtained ( with support of ODS/NIH) to also allow LC-isotope dilution MS determination. Our LC/MS method uses a C18 RP column with a gradient mobile phase. Detection was done with a photodiode array detector in sequence with a triple-quad mass spectrometer in the multiple reaction mode. Results for fortified foods by LC-UV/fluorescence and LC-MS are in good agreement. Ascorbic acid had to be stabilized by maintaining the sampling compartment at 4 degrees C to allow its simultaneous determination with the other WSV.
