Technical Abstract: Lignocellulosic biomass has the potential to supply many of our fuel and chemical feedstocks that are currently derived from fossil fuel sources. Xylan is one of the largest fractions of lignocellulose and must be hydrolyzed to simpler components to harness its full potential. We cultured microorganisms from samples collected from a hot springs environment that produced enzymes to break down xylan. We cloned a glycosyl hydrolase family 11 xylanase gene from a hot springs bacterium. The recombinant protein was expressed in Escherichia coli, purified, and biochemically characterized.
