Annual Entomological Society of Ammerica Meeting. Paper No. D0261. Technical Abstract: Diapause is the chief physiological mechanism enabling insect pests to survive and reproduce in a given area. Therefore, understanding the mechanism of diapause is critical in the management of any agroecosystem. In the Colorado potato beetle shifting of metabolism from development and reproduction to a metabolic profile enabling the beetles to survive a prolonged period of nonfeeding, anoxia and freezing temperatures occurs during the prediapause and early diapause phase. To clarify the mechanism(s) regulating diapause initiation, changes in lipid metabolism, respiration rates (overall metabolism) and gene expression were examined in prediapause and early diapause Colorado potato beetles. Three distinct observations were made: 1) The internal lipid quantity increases and the profile of external lipids changes over the prediapause period. 2) Respiration rates plateaus between days 4 and 8 and then decease to very low levels by day 15 post adult emergence. 3) The prediapause period is a time of dynamic changes in gene expression patterns, characterized by a shutdown of many metabolic genes between days 15 and 20 post emergence. The multiple competent analysis approach of this study demonstrates that the prediapause period is actually a sequence of separate developmental stages.
