Technical Abstract: In an effort to develop the molecular strategies to control rice (Oryzae sativa) diseases, molecular interactions of rice with rice blast [Magnaporthe oryzae, formerly (Magnaporthe grisea] and sheath blight (Rhizoctonia solani) fungi were analyzed. The interaction of rice with M. oryzae follows a biotrophic interaction at the early stage of infection whereas interaction of rice with R. solani follows a necrotrophic interaction. DNA microarrays and Serial Analysis of Gene Expression (SAGE) were used to analyze rice defense pathways. RNAs were prepared at 6, 10, 16 hrs after inoculation, gene expression profiles were compared. It was found that similar defense responses occurred 2 to 3 days earlier in the interactions of O. sativa /R. solani than the interaction of O. sativa/M. oryzae. However, unique differential genes expressed specific to O. sativa /M. oryzae and O. sativa /R. solani were also identified using real time PCR. These differences in gene expression are consistent with the differences in visible phenotypic reactions to the pathogen. The cause and effect of these defense genes in controlling rice fungal diseases will be discussed.
