Technical Abstract: Furanocoumarins in grapefruit juice have been shown to interfere with the oral bioavailability of medications primarily metabolized by intestinal CYP 3A4. The objective of this study was to isolate and structurally characterize furanocoumarins from grapefruit peel oil wax for in vitro structure/activity studies as well as for future small animal trials. Nonvolatile residues obtained from grapefruit peel oil were fractionated by flash chromatography, and subsequent isolations were achieved by preparative TLC and HPLC. Initial structural analyses were done by analytical HPLC-ESI-MS. Final structural elucidation was achieved by H1 and C13 NMR, FTIR, and high resolution MS. Initial HPLC-MS analyses of many of the compounds separated from the grapefruit nonvolatile residues shows the occurrence of compounds of widely diverse chemical structures. A number of these compounds have been successfully isolated, and their chemical structures and inhibition of the intestinal CYP 3A4 isozyme involved in grapefruit drug interactions will be reported.
