Technical Abstract: A major route of infection of avian influenza is through cells of the airway epithelium. To study the molecular mechanism of infection and early host responses we created a primary chicken tracheal cell culture. Epithelial cells were isolated from the trachea of 18 day old chicken embryos and cultured using an epithelial cell differentiation media. Positive immunostaining for cytokeratin, ciliar movement, and microarray comparison of gene expression with chicken fibroblast confirmed the epithelial in nature of the cells. In addition, the cells support replication and dissemination of lentogenic Avian Influenza (AI) and Newcastle disease virus (NDV) without any added protease.
