Technical Abstract: Bovine spongiform encephalopathy (BSE) is a fatal neurological disorder characterized by abnormal deposits of a protease-resistant isoform of the prion protein. In previous studies, we have characterized linkage disequilibrium (LD) and haplotype networks within the bovine prion gene (PRNP). Other studies have indicated possible associations of BSE with variation in the bovine PRND and PRNT genes located 16 kb and 26 kb 3’ to PRNP, respectively. The objective of the present study was to identify polymorphisms and define haplotypes in genomic region of bovine chromosome 13 containing PRND and PRNT. This will allow for 1) testing rare or common PRND or PRNT variation for association with BSE and 2) interpreting any association of PRND or PRNT alleles with BSE susceptibility. An approximately 18-kb genomic region containing PRND and PRNT was sequenced from 192 cattle representing the breadth of genetic diversity in U.S. beef and dairy cattle. LD and haplotypes defined by polymorphisms identified in this region will be presented.
