Technical Abstract: Marek's disease virus (MDV) is highly cell-associated virus. With no known receptors, it is of fundamental and applied interest to know how the virus is able to spread from cell to cell. Recently, we have evidence to suggest that MHC class II may be involved in the infection of T cells. First, R-LORF10 and LORF4 have been shown to interact in vitro with MHC class II invariant chain and b chain, respectively. Second, MDV-infected cells surprisingly up-regulate the surface expression of MHC class II. This suggests that a direct molecular mechanism for attracting and infecting T cells. To characterize the role of R-LORF10 and LORF4 in MHC class II expression, three recombinant MDVs derived from the MDV-BAC clone Md11gDloxP5-12 were generated. These viruses disrupted LORF4 (rMDVD LORF4), both copies (IRL and TRL) of R-LORF10 (rMDVD R-LORF10), or all genes (rMDVD LORF4DR-LORF10). The detailed data on MD pathogenesis of rMDVs in susceptible chickens will be discussed.
