Interpretive Summary:

Technical Abstract: We identified Congo red-binding (curli-producing) and -nonbinding phase variants of Escherichia coli O157:H7 strain ATCC 43895. The Congo red-binding variant, strain 43895OR, produced a red, dry and rough (rdar) phenotype on low-salt agar containing Congo red, and generated abundant biofilms on glass, polystyrene, and stainless steel. In contrast, variant 43895OW produced a smooth and white (saw) colony morphology and generated 5-8 fold less biofilm on the same surfaces. To better understand the genetic mechanisms involved in biofilm formation, we compared both variants using a variety of proteomic and genomic techniques. To date we have identified more than 20 proteins that are up-regulated in each variant during growth on low-salt agar. In addition, gene profiling experiments using reverse-transcriptase real-time PCR indicate that the regulation of biofilm formation in strain 43895OR is distinct from that previously described for other strain of E. coli.
