Technical Abstract: Bovine tuberculosis remains a costly disease in many countries despite extensive eradication and control efforts. Multiple wildlife reservoirs of Mycobacterium bovis infection found in recent decades in the US, Europe, New Zealand, and South Africa play important roles fuelling high rates of disease in the livestock. Innovative diagnostic tests for bovine tuberculosis are urgently needed to improve control strategies. A rapid antibody detection assay, VetTB STAT-PAK(TM), has been recently developed by Chembio Diagnostic Systems, Inc. (USA). This test uses the lateral-flow technology and a unique cocktail of recombinant antigens of M. bovis to detect specific antibodies of three immunoglobulin classes, IgG, IgM, and IgA, in cattle, deer, and other animal species. VetTB STAT-PAK(TM) kit is a simple and user-friendly assay that is easy to perform and read. It is stable at room temperature for up to 18 months, requires no laboratory equipment or skilled personnel. The test can use serum, plasma, or whole blood samples to provide result within 20 minutes. With serum samples from cattle in several countries, the diagnostic sensitivity of VetTB STAT-PAK(TM) kit varied from 65% in the UK to 83% in Nigeria. If used in conjunction with the intradermal skin test, the sensitivity of STAT-PAK(TM) kit could be boosted to over 90%. Specificity of VetTB STAT-PAK(TM) kit ranged from 94% to 100% in several experimental and field studies including samples from known M. avium reactors. Importantly, the specificity was not affected by the skin tests. STAT-PAK(TM) kits are proposed as a useful and cost-efficient ancillary assay for cattle and captive cervids. Also, this test will be most suitable for tuberculosis surveillance in a variety of wildlife and zoo species, especially if an instant result is needed.
