Interpretive Summary: Seed coat color, seed size, and seed shape of common bean largely defines consumer preference and market class. Thus, determination of the genetics of seed coat characteristics, such as color, pattern, and other demarcations, are important objectives in common bean breeding programs. Circumlineated (cl), is a seed coat trait that is identified by a physical groove in the seed surface at the boundary between seed coat colors. This study confirmed that Circumlineated is a recessive trait. In addition, molecular markers were developed that can be used to determine the genetic location of Circumlineated. This genetic information can be used to improve our knowledge about seed coat traits.
Technical Abstract: Circumlineated (cl) is identified by a precipitation line in the seed coat at the boundary of the white and colored zones. Circumlineated is recessive and is expressed in partly colored seed coats (t) with restricted patterns, such as virgarcus. The genetic location of Cl has not been confirmed, however molecular markers can be used to resolve its location. Early evidence for the genetic location of Cl indicated a weak linkage to T. In this study, AFLP and bulk segregant (BSA) analysis were used to identify co-segregating markers using Cl/Cl and cl/cl F3 progeny bulks from the cross t z cl G b v virgarcus BC3 5-593 x zsel Cl G b v sellatus BC3 5-593. One primer combination was found to yield unambiguous polymorphisms between the bulks of genotypes.
