Transmission of cucumber (Cucumis sativus) organelles is exceptional; chloroplast DNA shows maternal transmission while the mitochondrial (mt) DNA is exclusively paternally transmitted (J. Hered. 88:232-235). Transmission of plant organelles is sometimes under nuclear control. In cucumber, the Paternal Sorting of mt DNA (PSM) locus controls mt DNA sorting in plants possessing a mixture of wild-type and mutated (MSC) mt DNAs. The wild-type (+) allele at PSM conditions paternal transmission of wild-type or mutated mitochondria. The (-) allele at PSM preferentially selects for rare wild-type sublimons present in cucumbers possessing MSC mitochondria (J. Hered. 95:492-497). We placed the PSM locus onto the genetic map of cucumber using an F2 family from the cross of Plant Introduction (PI) 401723 with Straight 8 (ST8). Each F2 plant was pollinated with the mosaic cucumber MSC16 and progenies scored as all mosaic (Psm+/Psm+), all wild-type (Psm-/Psm-), or 1:1 wild-type to mosaic (Psm+/Psm-). Amplified fragments length polymorphisms (AFLP) were identified closely linked to PSM and were then placed on the cucumber genetic map. The identification and mapping of genetic markers closely linked to PSM will eventually allow for the cloning of this unique locus that controls sorting of paternally transmitted mt DNAs and provide insights about nuclear control of the sorting of organellar DNAs in plants.
