Interpretive Summary:

Technical Abstract: Chlorotic fleck is a strawberry disease of unknown etiology for which the causal agent has been transmitted with the cotton aphid and speculated to be a virus. In the course of a project that involved the identification of causal agents for all strawberry diseases of unknown etiology, a chlorotic fleck infected plant was acquired from the National Clonal Germplasm Repository in Corvallis, Oregon. Double-stranded RNA was extracted and cloned. The plant was infected with both pallidosis agents, in addition to a novel virus, designated as Strawberry chlorotic fleck associated virus, with close homology to members of the genus Closterovirus, the aphid-borne members of the Closteroviridae. The majority of the genomic sequence obtained reveals a unique genome organization with features of both Beet yellows virus (BYV) and Citrus tristeza virus (CTV). Phylogenetic analysis indicates the close relationship of the virus, with CTV and BYV. A molecular detection test has been developed while transmission tests are under way to determine the aphid species able to transmit the virus.
