Technical Abstract: The analysis of carbon isotopomers by mass spectrometry and 13C-NMR is a powerful tool for assessing metabolic turnover. Combining this approach with glycomics enables metabolic flux analysis on a global scale (fluxomics), and provides a large number of criteria with which to assess biological perturbations. The aim of this technique is to quantify the fractional incorporation of **13C from exogenous **13C-labeled precursors into all cellular carbohydrates. Tracing carbohydrate metabolic pathways by LC-electrospray-MS, GC-EI-MS, and HSQC-NMR, and new MS techniques using diethyldithioacetal acetates to estimate **13C-flux into the characteristic anomeric carbon-1 of aldoses will be described. The potential of chirally-deuterated (S)-[6'-**2H1]glucose as a tracer will also be discussed. Our current interest includes glycomic flux in human salivary glycoproteins, the bifido pathway in lactic acid bacteria, and biosynthesis of the 11-carbon amino-dialdose, tunicamine.
