Technical Abstract: Two objectives of this NSF-funded project are to generate and map expressed sequence tags (ESTs) to wheat chromosomes using deletion lines. This report summarizes the results of ESTs that have been mapped on group 1 chromosomes (1A, 1B, and 1D). As of August 21, 2002, a total of 1,394 EST loci derived from 560 probes were mapped to the group 1 chromosomes. This number was higher than the average number of ESTs mapped per chromosome group (1,141). The distribution of mapped ESTs among the chromosomes was significantly different (P<0.01) with 426, 529, and 439, for 1A, 1B, and 1D, respectively. The arm difference of mapped ESTs was also highly significant (P<0.01) for all the three chromosomes with larger numbers of ESTs on the long arm than the short one. The distribution of ESTs along the chromosome arms was found to be non-random, with the occurrence of clustering in all six chromosome arms and the satellite region of 1BS highly significant at P<0.01. ESTs were generally clustered in the distal regions and had lower density in the proximal regions. However, a difference existed among the three chromosomes for the chromosomal distribution pattern of EST loci even though they are believed to be highly homologous. The number of ESTs mapped in the terminal area exceeded the expected for chromosome arms 1AS, 1DS, 1DL and 1BS-sat. For chromosome arms 1AL, 1BS, and 1BL, the total number of ESTs mapped in the terminal region was equal to or less than the expected number. This difference of gene distribution pattern among and along the chromosomes may reflect, to some extent, the functional differentiation of homologous chromosomes in the long-term process of evolution in the tribe Triticeae. An EST consensus map of group 1 chromosomes, and relationship of gene function with the chromosomal distribution pattern will also be reported.
