Technical Abstract: Septoria tritici blotch caused by Mycosphaerella graminicola is a serious fungal disease of wheat worldwide. Developing wheat cultivars resistant to M. graminicola is complicated by a long latent period, environmental effects on symptom expression, and variation among evaluators in the use of disease scoring systems. To circumvent these problems, we developed a fluorescent real-time PCR assay to quantify the biomass of M. graminicola in wheat cultivars. A set of specific primers was designed from B-tubulin gene sequences, which amplified a PCR product of 100 bp from pure cultures and infected wheat leaves only. The real-time PCR analysis showed that the biomass of M. graminicola and disease severity in the resistant cultivars, Tadinia and W7984, were significantly less than those in either of the susceptible checks, Yecoro Rojo and Opata85, beginning 12 d after inoculation. Real-time PCR appears to be a valuable tool for discriminating between resistant and susceptible wheat.
