Technical Abstract: This NRSP008 National Animal Genome Research Program station report documents progress at Beltsville on swine genome efforts. The BARC research has focused on functional genomics. With collaborators at Baylor Univ. in Texas, BARC scientists and members of the international SLA nomenclature committee developed new molecular nomenclature and phylogenies for swine Major Histocompatibility Complex (MHC) or Swine Leukocyte Antigen (SLA) class I genes and haplotypes. Future work will extend this molecular nomenclature and phylogenies for SLA class II genes. With collaborators at the Nutrient Requirements & Functions Lab, Beltsville Human Nutrition Research Center, they have expanded real-time expression assays for panels of immune markers known to control vaccine and disease immunity (>350 genes targeted; real-time assays now available for >200 genes). Established patterns of gene expression during innate and adaptive [T helper 1 (Th1) and Th2] immune responses have been characterized with tissues collected from infected pigs. Focus is now directed toward upstream regulatory sequences. Future uses will target gene expression patterns for understanding protective immunity to respiratory infections, e.g., bacterial infections such as Mycoplasma hyopneumoniae, and viral infections, porcine reproductive and respiratory syndrome virus (PRRSV), in swine.
