Technical Abstract: DNA marker technology is being used to assist in the breeding of turfgrasses. SSR markers are valuable because they are codominant, portable, and PCR-based. In this study, SSRs were isolated from four grasses: Kentucky bluegrass (Poa pratensis L.), Texas bluegrass (Poa arachnifera Torr.), zoysiagrass (Zoysia matrella L.), and a bermudagrass hybrid [Cynodon dactylon (L.) Pers. x C. transvaalensis Burtt-Davy]. For each species and the hybrid, the following steps were conducted: 1) a small insert genomic library was developed, 2) plasmid DNA was isolated and sequenced, 3) SSR primers were designed from sequence data, and 4) SSRs were surveyed in genotypes from each of the respective species. These SSRs have potential application across these three genera as well as other turf and forage grasses. The use of these SSRs in characterizing genetic diversity and conducting mapping studies will be discussed.
