Interpretive Summary: The development of genetic maps can aid in the improvement of crops for insect and disease resistance and also speed progress in developing new varieties with improved flavor, nutritional value and storage life. A genetic map for peach has been developed. This map is available in a public database and can be used by peach breeders and other scientists who work on the improvement of peaches, nectarines and related fruits such as plums, apricots, and cherries.

Technical Abstract: Peach (Prunus persica) represents a model species for the Rosaceae, which includes a number of economically important fruit trees. To develop an extensive Prunus EST database for identification and cloning of genes important to fruit and tree development, we generated 9984 high-quality expressed sequence tags (ESTs) from a peach cDNA library of developing fruit mesocarp. After assembly and annotation, a putative peach unigene set consisting of 3842 ESTs was defined. Gene Ontology (GO) classification was assigned based on the annotation of the single "best hit" match against SWISS-PROT database. No similar sequences could be found in the databases for about 24% of the sequences, which might represent fruit tree specific sequences. Using core markers from the general Prunus genetic map, BAC clones were anchored on the genetic map providing a framework for construction of a physical and a transcript map. To develop a transcript map, 1236 ESTs from the putative peach unigene set and additional 68 peach cDNA clones were hybridized against the peach BAC library. Hybridizing ESTs to genetically anchored BACs immediately localized 11.2% of the ESTs on the genetic map. ESTs showed a clustering of expressed genes in defined regions of the linkage groups.
