Interpretive Summary:

Technical Abstract: In-vivo, in-vitro and reporter gene systems have previously shown induction of the selenoprotein TR by SF purified from broccoli. This upregulation is mediated by the antioxidant response element (ARE) found in the promoter of the gene. The present study examined the nutritional interaction between deficient or supranutritional amounts of dietary Se and SF on regulation of TR, GPX and QR. In a 2X2 factorial design (N=8/TRT), rats were fed deficient or supranutritional Se (0.05 or 2.0 mg Se/kg diet as selenite) diets for 32 d, and were gavaged with 500 uMol SF/kg-1 or carrier daily for the last 5 d of the study. These data suggest that GPX is only responsive to dietary Se and probably not regulated by an ARE while TR is regulated by an ARE and is responsive to both Se and SF. The effect of SF and Se on TR, QR and GPX was tissue dependant, but it is not known whether this is related to SF or Se availability to the tissues, or to tissue specific transcription factor expression.
