Interpretive Summary: Potyviruses routinely cause significant yield and quality losses in cucumber and melon. Sources of ZYMV resistances have been identified in both crops, however they show different modes of inheritance and their genetic relationships are unknown. We compared the genetic location of ZYMV resistances in cucumber and melon to that different regions condition these resistances, indicating that we cannot use genetic markers developed in one crop to select for ZYMV resistances in the other crop. This research will be of interest to breeders and pathologists working to develop potyvirus resistant cucurbit cultivars.

Technical Abstract: Potyviruses routinely cause significant losses in the cucurbits. Sources of ZYMV resistances have been identified and mapped in cucumber (Cucumis sativus L.) and melon (Cucumis melo L.); however they show different modes of inheritance and their genetic relationships are unknown. Comparative mapping of ZYMV resistance loci was conducted using the simple sequence repeat (SSR) CMAG36 linked to the Zym-1 locus of melon and a genomic region cosegregating with the zym locus of cucumber. The CMAG36 homologous region was cloned and sequenced from cucumber and an RFLP identified, mapped in cucumber, and segregated independently from the zym locus. A 5-kb genomic region (YCZ-5) carrying an AFLP cosegregating with resistance to ZYMV in cucumber was cloned and sequenced. This genomic region possessed SNPs and small insertions or deletions distinguishing alleles from the ZYMV-resistant (TMG1) and susceptible (Straight 8) cucumbers; however all evaluated polymorphisms were unique to the ZYMV-susceptible parent (Straight 8). The cucumber YCZ-5 region was hybridized to melon bacterial artificial chromosome (BAC) clones and a 180-kb contig assembled. One end of the melon contig was mapped in cucumber and cosegregated with ZYMV resistance, demonstrating colinearity across this region between the physical map of melon and the genetic map of cucumber. This melon region was mapped and segregated independently of ZYMV resistance loci in two families of melon, indicating that sources of ZYMV resistances from cucumber TMG1 and melon PI 414723 are non-syntenic.
