Interpretive Summary: Myogenic factor 6 (MYF6) is an important gene in muscle development. MYF6 and probably MYF4 appear to be involved in the development of myotubes and mutations in the human MYF6 are associated with myopathy and the severe condition of Becker muscular dystrophy. This important gene was mapped to the end of pig chromosome 5 by two independent methods. Our results of linkage and RH mapping are in agreement with comparative mapping data. This gene could be associated with differences in muscle mass of market pigs.

Technical Abstract: A fragment of the porcine MYF6 (myogenic factor 6; herculin), gene encompassing parts of exons 1 and 2 and the intervening intron was amplified. To verify the identity, the 379 bp amplicon was directly sequenced and the sequence data matched exon 1 of the human sequence with identity of 90% (64/70 nucleotides). In intron 1 of the MYF6 amplimer three SNP were identified by means of BseRI, AvaI and AcyI restriction digests caused by G/C, C/T and C/A polymorphisms, respectively. Codominant inheritance of alleles at all three loci was confirmed in the USDA-MARC reference pedigree. Allele frequencies at these loci of MYF6 were determined in eight pig breeds. Multiple-point linkage analysis of the USDA-MARC swine genotypic data based on 27 informative meioses for MYF6 placed the MYF6 gene at position 146 cM on the updated USDA-MARC map (www.marc.usda.gov). Radiation hybrid (RH) mapping was performed with the use of the INRA-University of Minnesota porcine Radiation Hybrid panel. Two-point analysis performed on IMpRH Server at http:/imprh.tolouse.inra.fr showed the most significant linkage to SW967 (LOD = 13.31, 31 cR), which is the most distal marker on q arm of chromosome 5, and to the nearby marker SW378 (LOD = 6.41, 61 cR) on the first porcine RH map.
