Technical Abstract: The temperature sensitivity of chlorophyll accumulation was used to identify acquired thermotolerance deficient mutants in Arabidopsis. This study describes the fine mapping of acquired thermotolerance deficient mutants of RLD, Columbia and WS2 ecotypes. The RLD and Columbia acquired thermotolerance deficient mutants were selected by screening M2 populations of Arabidopsis thaliana seedlings derived from seeds treated with the mutagen ethyl methane sulfonate. Etiolated seedlings preincubated at 38C for 4 h to induce thermotolerance and then challenged at 44C for 30 min in the dark turned green with 24 h when exposed to continuous light. The mutant seedlings were light green with 24 h, but attained full Chl levels within 72 h. This delayed pattern of chlorophyll accumulation was observed in control seedlings challenged at 44C that had not been exposed to the 38C preincubation. Mapping populations were evaluated with PCR to score the co-segregation of the mutant phenotype with simple sequence-length polymorphic markers. Chromosomal locations of the mutated genes were further targeted by evaluating co-segregation of the mutant phenotype with single nucleotide polymorphisms. Mapping of our AtTS02 acquired thermotolerance deficient mutant derived from a RLD ecotype revealed that the mutation is localized to a region of Chromosome 3 devoid of heat shock proteins.
