Title: Genome-Wide Physical Mapping of Pathogen-Response Genes in Barley

Technical Abstract: Genes involved in resistance to pathogens are an intricated network of cell signaling events. A genome-wide search was undertaken to identify gene-rich BAC clones from barley that contained defense-related genes. HarvEST Triticeae (http://harvest.ucr.edu/) was utilized to identify ESTs uniquely associated with pathogen challenge. Of the 271,317 sequences in HarvEST v 0.90, 1,372 contigs and 3,492 singletons (4,864 total) are exclusive to barley-Blumeria or barley-Fusarium interactions, and not represented anywhere else in the 271,317 ESTs. Two-hundred of these ESTs were used to design overgo probes to hybridize to the barley BAC library for the purpose of understanding the distribution of pathogen response genes and facilitate gene cloning. 3,678 hybridizing clones at a 1x genome equivalent of 2.92 BACs/overgo were identified. Among the 3,678 clones, 108 addresses hybridized to more than one overgo pool. These 108 clones are being fingerprinted and mapped using FPC to determine if there are physically linked, defense-related gene clusters in the barley genome.
