Interpretive Summary:

Technical Abstract: Quantitation of genes involved in regulating disease resistance in swine is essential for identifying pigs, which are genetically resistant to infections. To this end we have developed quantitative TaqMan real-time PCR assays for assessing mRNA levels of immune regulatory molecules, specifically for porcine cytokines, including IFN-?, IL-4, IL-5, IL-10, IL-12 p35/p40, IL-15, IL18, and receptors, and the housekeeping genes, rRNA and HPRT. The assays were validated by 1) use of specific primer and probe sequences, 2) comparison of results with semiquantitative RT-PCR, and 3) expression in tissues from pigs infected with the parasites, Toxoplasma gondii or Ascaris suum. These assays will be used to compare immune phenotypes of lines of pigs to determine responses at different developmental stages and after vaccinations and infections.
