Technical Abstract: The production of mannitol by fermentation and/or enzymatic means has become attractive because of the problems associated with its production chemically. Mannitol is a naturally occurring polyol widely used in the food, medicine, pharmaceutical, and chemical industries. In this presentation, the production of mannitol from fructose by Lactobacillus intermedius NRRL B-3693 (selected after screening 72 bacterial cultures from ARS Culture Collection) will be described. The bacterium produced mannitol, lactic acid, and acetic acid from fructose, fructose/glucose (2:1), or fructose/sucrose (1:2) co-substrate mixtures. Glucose can be replaced by maltose, galactose, mannose, or starch with glucoamylase. It produced lactic acid and ethanol but no acetic acid from glucose. Factors affecting the production of mannitol and lactic acid by the bacterium in pH-controlled batch and fed-batch fermentations will be presented. Current status of microbial production of mannitol, problems and prospects of mannitol production by heterofermentative lactic acid bacteria, and future directions of research will be outlined.
