Technical Abstract: Research efforts in the past two decades have revealed the complex mechanisms employed by fungi to control gene activity. The tremendous expansion in our knowledge of the regulation of nitrogen metabolism and carbon metabolism, due largely to the powerful combination of genetics, biochemistry, and molecular biology approaches, provides us the framework that would allow us to test many of the long-standing hypotheses about how gene activity is regulated. This article reviews the roles of global, pathway-specific and repressor proteins, and the functional interplay of these factors in modulating gene activity involved in the utilization of alternative nitrogen and carbon sources. The review also emphasizes recent progress in the characterization of fungal genes with novel regulatory functions and affecting nitrogen and carbon metabolism directly or indirectly.
