Title: Vector-Based Homology Analysis of Data Sets from Diverse Taxa for a Triticeae Microarray

Technical Abstract: Rapid identification of agronomically important genes is a focal point of microarray technology. We are utilizing emerging EST resources to initiate a gene discovery program targeted at biotic stress in barley. A unigene set was derived from 63,802 CUGI barley ESTs representing 13 distinct cDNA libraries. The Arabidopsis pathoarray and various crop databases will be utilized to rapidly identify barley homologs that are important in resistance signaling. By using a program that can convert DNA sequences into real-valued vectors, the angles between them can be used to determine homology. This vector based technology is a standard pattern recognition technique that functions rapidly and efficiently. Non-unique sequences will be masked by comparison to a Triticeae homology and repetitive element database. This information will be used to design 60-mers for an oligo-based microarray.
