Title: Identification of Cdna Markers Linked to Hessian Fly Resistance Gene H17 in Wheat

Technical Abstract: The Hessian fly (Mayetiola destructor (Say)) is one of the most destructive pests of wheat in many areas of the United States. Genetic interactions between the Hessian fly and Triticum follow a gene-for-gene relationship. At least 26 genes for resistance to the Hessian fly have been identified in various Triticum species and transferred into common wheat (T. aestivum L.). The resistance gene H17 was identified in durum wheat (T. turgidum L.) and confers resistance to all Hessian fly biotypes that have been tested. By traditional genetic analysis, H17 was mapped to chromosome 5A. Nonetheless, fine mapping of H17 through molecular marker analysis will not only facilitate its application in marker-assisted breeding programs but also will be helpful for the isolation of H17 through a map-based cloning strategy. Our objective in this research was to identify cDNA markers closely linked to H17. cDNA markers are believed to be the most suitable for comparative mapping between wheat and rice. For this purpose, total RNA from a pair of near isogenic lines (NILs), Newton and H17 in a Newton background, was used for mRNA differential display. Forty eight primer combinations were used for PCR amplification. In addition, a subtractive cDNA library from the H17 line was constructed. Several differentially expressed cDNAs were identified. The linkage relationship between these cDNA fragments and H17 will be clarified.
