Technical Abstract: Talaromyces flavus (anamorph = Penicillium dangeardii) is a soil-inhabiting fungus that controls diseases caused by Verticillium spp. This study was undertaken to determine the genetic diversity of T. flavus and other species of Talaromyces, as well as to determine the relatedness of T. flavus to a published ITS sequence for a human pathogen, Penicillium marneffei. Talaromyces spp. were from 18 countries (31 isolates) and from 22 states in the US (29 isolates). Authentic cultures of T. emersonii, T. helicus, T. trachyspermus, and T. wortmannii were obtained from ATCC. Growth rate of each strain at 22 and 37 C was determined. DNA was extracted from all strains and the ITS1 and ITS2 regions, as well as the gpd (glyceraldehyde-3-phosphate dehydrogenase) region, were amplified and sequenced. There was less variability in ITS regions than in the gpd region. Correlation between dendrograms for ITS and gpd sequences was poor, as was correlation between ITS and gpd sequences and gross morphology. Dendrogram using ITS sequences indicated that there were 2 major groups. The larger ITS group contained 38 isolates, including T. flavus Tf-1 and another biocontrol strain.
